The first train usually departs around 7:00 AM, and the last one around 8:00 PM. The journey from Nagoya to Matsumoto takes about 2 hours and 10 minutes. JR West Timetable and Fare Finder has all the schedules. I believe tickets can be reserved on the JR West site: https://www.westjr.co.jp/global/en/ticket/overview/ Reserved tickets can only be picked up at these stations: https://www.jr-odekake.net/eki/global/en/howto/train-reservation/receive/station There are other third party reservations sites online that offer reservations, google "shinano express reservations". If you are going to arrive in Japan a few days before your Nakasendo trail trip you can buy a ticket at a JR ticket office.
